Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate YWHAZ in samples. An antibody specific for YWHAZ has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyYWHAZ present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for YWHAZ is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of YWHAZ bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:YWHAz belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse, rat and sheep orthologs.
The deduced 245-amino acid protein shares high sequence homology with other 14-3-3 subtypes. The zeta mRNA was widely expressed in various gray matter brain regions, including the neocortex, hippocampus, caudate-putamen, thalamus, cerebellar cortex, and several brain stem nuclei. A human protein with phospholipase A2 activity was shown to be the zeta subtype of the 14-3-3 protein
