Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate VCAM1/CD106 in samples. An antibody specific for VCAM1/CD106 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVCAM1/CD106 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VCAM1/CD106 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VCAM1/CD106 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:VCAM-1 (CD106), a member of the immunoglobulin superfamily, is a cell surface protein expressed by activated endothelial cells and certain leukocytes (such as macrophages). VCAM-1 expression is induced by IL-1β, IL-4, TNF-α and IFN-γ. VCAM-1 binds to leukocyte integrins VLA-4 and α4β7. The rat and mouse SVCAM-1 proteins share approximately 76% amino acid similarity.
VCAM-1 is a member of the Ig superfamily and encodes a cell surface sialoglycoprotein expressed by cytokine-activated endothelium. This type I membrane protein mediates leukocyte-endothelial cell adhesion and signal transduction, and may play a role in the development of artherosclerosis and rheumatoid arthritis.
