Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate UGCG in samples. An antibody specific for UGCG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUGCG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UGCG is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UGCG bound in the initial step. The color development is stopped and the intensity of the color is measured.

The assembled full-length HUGT1 cDNA encodes a deduced 1,555-amino acid protein with a predicted ER-targeting signal sequence, ER retrieval signal, 3 N-glycosylation sites, and 2 disulfide loops. HUGT1 shares 55% sequence identity with HUGT2 and 31 to 45% identity with Drosophila, C. elegans, and S. pombe homologs. Northern blot analysis detected 3 HUGT1 transcripts of 5.7, 8, and 10.6 kb in pancreas, heart, brain, placenta, lung, liver, skeletal muscle, and kidney.
