Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate TRH in samples. An antibody specific for TRH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTRH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TRH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TRH bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Thyrotropin releasing hormone, a hormone secreted by the hypothalamus that stimulates release of thyrotropin. TRH can also be detected in other areas of the body including the gastrointestinal system and pancreatic islets. Thyroid-stimulating hormone, also known as thyrotropin, is secreted from cells in the anterior pituitary called thyrotrophs. The most important controller of TSH secretion is thyroid-releasing hormone.
TRH is produced by the hypothalamus in medial neurons of the paraventricular nucleus. Initially, it is synthesized as a 242 amino acid precursor polypeptide that contains 6 copies of the sequence -Gln-His-Pro-Gly-, flanked by di-basic peptides that are later processed through proteolysis to give the mature TRH molecule.
