Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate TPPP in samples. An antibody specific for TPPP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTPPP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TPPP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TPPP bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:The novel basic, heat-stable tubulin polymerization promoting protein TPPP/p25 is associated with microtubules in vitro and can induce the formation of aberrant microtubule assemblies.
Immunohistochemistry and confocal microscopy demonstrates that TPPP/p25 is enriched in filamentous alpha-synuclein bearing Lewy bodies of Parkinson's (PD) and diffuse Lewy body disease (DLBD), as well as glial inclusions of multiple system atrophy (MSA). There is a correlation between TPPP/p25 and alpha-synuclein immunoreactivity in Western blot. In contrast, TPPP/p25 is not associated with abnormally phosphorylated tau in various inclusions of Pick's disease (PiD), progressive supranuclear palsy (PSP), and corticobasal degeneration (CBD).
