Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate STAT6 in samples. An antibody specific for STAT6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySTAT6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for STAT6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of STAT6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:STAT6 is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein plays a central role in exerting IL4 mediated biological responses.
It is found to induce the expression of BCL2L1/BCL-X(L), which is responsible for the anti-apoptotic activity of IL4. Knockout studies in mice suggested the roles of this gene in differentiation of T helper 2 (Th2) cells, expression of cell surface markers, and class switch of immunoglobulins.
