Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate ADAM22 in samples. An antibody specific for ADAM22 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADAM22 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAM22 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAM22 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:ADAM22 is a member of the ADAM family. Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis.
Three-prime RACE yielded products corresponding to 2 MDC2 mRNAs, MDC2-alpha and -beta, in both mouse and human. MDC2-alpha encodes a predicted 859-amino acid protein. The shorter transcript, MDC2-beta, results from exon skipping and encodes an 823-amino acid protein. The MDC2-alpha protein shares 56% and 51% sequence similarity with MDC and MDC3, respectively.
