Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate ADCY2 in samples. An antibody specific for ADCY2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADCY2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADCY2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADCY2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Adenylate cyclase type 2 is a member of the family of adenylate cyclases, which are membrane-associated enzymes that catalyze the formation of the secondary messenger cyclic adenosine monophosphate (cAMP). This enzyme is insensitive to Ca(2+)/calmodulin, and is stimulated by the G protein beta and gamma subunit complex.
The amino acid sequence of ADCY2 displayed significant homology with ADCY8 in the highly conserved adenylyl cyclase domain (250 amino acids) found in the 3-prime cytoplasmic portion of all mammalian adenylyl cyclases. However, outside this domain, the homology was extremely low.Semiquantitative RT-PCR detected ADCY2 in all tissues examined except peripheral blood leukocytes. Highest expression was detected in brain, skeletal muscle, and testis.
