Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PYCR2 in samples. An antibody specific for PYCR2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPYCR2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PYCR2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PYCR2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:PYCR2, Belongs to the pyrroline-5-carboxylate reductase family. Pyrroline-5-carboxylate reductase(P5CR) is the enzyme that catalyzes the terminal step in the biosynthesis of proline from glutamate, the NAD(P) dependent oxidation of 1-pyrroline-5-carboxylate into proline.
Pyrroline-5-carboxylate reductase, which converts pyrroline-5-carboxylate to proline, has been identified in human erythrocytes. The level of pyrroline-5-carboxylate reductase activity in these cells is comparable to the activity levels of major erythrocyte enzymes. The physiologic function of the enzyme in erythrocytes cannot be related to its function in other tissues, i.e., producing proline for protein synthesis.
