Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PTHLH in samples. An antibody specific for PTHLH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPTHLH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTHLH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTHLH bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Parathyroid hormone-related protein (or PTHrP) is a protein occasionally secreted by cancer cells (breast cancer, certain types of lung cancer including squamous cell carcinoma). However, it also has normal functions. PTHrP acts as an endocrine, autocrine/ paracrine, and intracrine hormone. PTHrP is related in function to the "normal" parathyroid hormone. When a tumor secretes PTHrP, this can lead to hypercalcemia. As this is sometimes the first sign of the malignancy, hypercalcemia caused by PTHrP is considered a paraneoplastic phenomenon. PTHrP shares the same N-terminal end as parathyroid hormone and therefore it can bind to the same receptor (the Type I PTH receptor). PTHrP aids in normal mammary gland development and lactation as well as placental transfer of calcium.
