Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PRX in samples. An antibody specific for PRX has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRX present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRX is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRX bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Periaxin is a protein involved in peripheral nerve myelin upkeep. The encoded protein contains 2 PDZ domains which were named after PSD95 (post synaptic density protein), DlgA (Drosophila disc large tumor suppressor), and ZO1 (a mammalian tight junction protein). Two alternatively spliced transcript variants have been described for this gene which encode different protein isoforms and which are targeted differently in the Schwann cell. Mutations in this gene cause Charcot-Marie-Tooth neuoropathy, type 4F and Dejerine-Sottas neuropathy. The PDZ domain, which consists of a nearly 90-amino acid protein-binding motif that interacts with the C termini of plasma membrane proteins and with the cortical cytoskeleton, has been implicated in the assembly of signaling complexes at sites of cell-cell contact.
