Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PDPN in samples. An antibody specific for PDPN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPDPN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDPN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDPN bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Podoplanin is a type-I integral membrane glycoprotein with diverse distribution in human tissues. The physiological function of this protein may be related to its mucin-type character. The homologous protein in other species has been described as a differentiation antigen and influenza-virus receptor. The specific function of this protein has not been determined but it has been proposed as a marker of lung injury. Alternatively spliced transcript variants encoding different isoforms have been identified.Highly expressed in placenta, lung, skeletal muscle and brain. Weakly expressed in brain, kidney and liver. In placenta, expressed on the apical plasma membrane of endothelium. In lung, expressed in alveolar epithelium. Up-regulated in colorectal tumors and expressed in 25% of early oral squamous cell carcinomas.
