Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PDGFRL in samples. An antibody specific for PDGFRL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPDGFRL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDGFRL is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDGFRL bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:PDGFRL is a protein with significant sequence similarity to the ligand binding domain of platelet-derived growth factor receptor beta. Mutations in this gene, or deletion of a chromosomal segment containing this gene, are associated with sporadic hepatocellular carcinomas, colorectal cancers, and non-small cell lung cancers. This suggests this gene product may function as a tumor suppressor.
The 375-amino acid protein predicted from this cDNA has hydrophobic sequence at the NH2 terminus, characteristic of signal peptides. It shows 27% and 25% identity with the extracellular domain of platelet-derived growth factor receptor-beta and FMS-like tyrosine kinase (FLT), respectively.
