Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate PADI3 in samples. An antibody specific for PADI3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPADI3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PADI3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PADI3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Peptidyl arginine deiminase, type III, also known as PADI3, is a member of the peptidyl arginine deiminase family of enzymes, which catalyze the post-translational deimination of proteins by converting arginine residues into citrullines in the presence of calcium ions. The family members have distinct substrate specificities and tissue-specific expression patterns.
The type III enzyme modulates hair structural proteins, such as filaggrin in the hair follicle and trichohyalin in the inner root sheath, during hair follicle formation. Together with the type I enzyme, this enzyme may also play a role in terminal differentiation of the epidermis.
