Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate ADAMTS13 in samples. An antibody specific for ADAMTS13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADAMTS13 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAMTS13 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAMTS13 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:ADAMTS13 (a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13)also known as von Willebrand factor-cleaving protease (VWFCP)is a zinc-containing metalloprotease enzyme that cleaves von Willebrand factor (vWf), a large protein involved in blood clotting. It is secreted in blood and degrades large vWf multimers, decreasing their activity.Genomically, ADAMTS13 shares many properties with the 19 member ADAMTS family, all of which are characterised by a protease domain (the part that performs the protein hydrolysis), an adjacent disintegrin domain and one or more thrombospondin domains. ADAMTS13 in fact has eight thrombospondin domains. It has no hydrophobic transmembrane domain, and hence it not anchored in the cell membrane.
