Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate LGALS9 in samples. An antibody specific for LGALS9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLGALS9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LGALS9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LGALS9 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Gal-9 was first cloned from tumor tissues of Hodgkins disease that is frequently characterized by eosinophilia. Galectins exhibit various biological functions, such as cell aggregation and adhesion, proliferation,cell death, modulation of inflammation and metastasis.First, Gal-9/ecalectin was identified as a potent T cellderived eosinophil chemoattractant (ECA).
Gal-9 exhibits ECA activity in vitro and in vivo, activates eosinophils, and influences eosinophil survival. It requires divalent galactoside- binding activity but not its linker peptide to exhibit its functions , and some matrix metalloproteinases are involved in the release.
