Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate KLK8 in samples. An antibody specific for KLK8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKLK8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KLK8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KLK8 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Originally described from mouse brain, KLK8 was implicated in neural plasticity and learning. Kallikrein 8 is thought to be involved in neurite outgrowth and fasciculation during nervous system development. Kallikrein 8 is induced in neurite injury, and Kallikrein 8 has been shown to be elevated in the hippocampus on Alzheimers patients.
Kallikrein 8 is thought to regulate the early phase of long-term potentiation (LTP), possibly by cleaving fibronectin and other substrates.Kallikrein 8 is also found in the uterus, where it is hormonally regulated. Cervical cancer cell lines express high levels of Kallikrein 8, and it may be useful as a tumor marker. Kallikrein 8 is not limited to the brain and uterus, as many tissues and cell lines make the protein.
