Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate DSG2 in samples. An antibody specific for DSG2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDSG2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DSG2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DSG2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Desmosomes are cell-cell junctions between epithelial, myocardial, and certain other cell types. Desmoglein 2 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Currently, three desmoglein subfamily members have been identified and all are members of the cadherin cell adhesion molecule superfamily. These desmoglein gene family members are located in a cluster on chromosome 18.
Desmoglein-2 is expressed in colon, colon carcinoma, and other simple and stratified epithelial-derived cell lines. Mutations in this gene have been associated with arrhythmogenic right ventricular dysplasia, familial, 10.
