Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate DLG4 in samples. An antibody specific for DLG4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDLG4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DLG4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DLG4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Disks large homolog 4 is a member of the membrane-associated guanylate kinase (MAGUK) family. It heteromultimerizes with DLG2. With DLG2 it is recruited into the same NMDA receptor and potassium channel clusters. These two MAGUK proteins may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins. DLG4 is the best studied member of the MAGUK-family of PDZ domain-containing proteins. Like all MAGUK-family proteins, its basic structure includes three PDZ domains, an SH3 domain, and a guanylate kinase-like domain (GK) connected by disordered linker regions. It is almost exclusively located in the post synaptic density of neurons, and is involved in anchoring synaptic proteins.
