Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate OMD in samples. An antibody specific for OMD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyOMD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for OMD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of OMD bound in the initial step. The color development is stopped and the intensity of the color is measured.

ORM is an acute phase (Acute phase protein) plasma alpha-globulin glycoprotein and is modulated by two polymorphic genes. It is synthesized primarily in hepatocytes and has a normal plasma concentration between 0.6-1.2 mg/mL (1-3% plasma protein).Plasma levels are affected by pregnancy, certain drugs, and certain diseases, particularly HIV. The only established function of ORM is to act as a carrier of basic and neutrally charged lipophilic compounds. In medicine, it is known as the primary carrier of basic drugs (whereas albumin carries acidic drugs), steroids, and protease inhibitors.Orosomucoid increases in amount in obstructive jaundices while diminishes in hepatocellular jaundice and in intestinal infections.
