Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate SERPINA3 in samples. An antibody specific for SERPINA3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySERPINA3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SERPINA3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SERPINA3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Alpha 1-antichymotrypsin is an alpha globulin glycoprotein that is a member of the serine proteinase inhibitor (serpin) family.It inhibits the activity of certain enzymes called proteinases, such as cathepsin G that is found in neutrophils, and chymases found in mast cells, by cleaving them into a different shape or conformation. This activity protects some tissues, such as the lower respiratory tract, from damage caused by proteolytic enzymes.
This enzyme is produced in the liver, and is an acute phase protein that is induced during inflammation. Deficiency of this protein has been associated with liver disease. Mutations have been identified in patients with Parkinson disease and chronic obstructive pulmonary disease.
