Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate GEMIN6 in samples. An antibody specific for GEMIN6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGEMIN6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GEMIN6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GEMIN6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:GEMIN6 is part of a large macromolecular complex, localized to both the cytoplasm and the nucleus, that plays a role in the cytoplasmic assembly of small nuclear ribonucleoproteins (snRNPs). Other members of this complex include SMN , GEMIN2 (SIP1), GEMIN3 (DDX20), GEMIN4, and GEMIN5.
The deduced 167-amino acid protein has a predicted molecular mass of 18 kD and shares 86% sequence identity with the mouse homolog. GEMIN6 migrates as a 16-kD protein by Western blot analysis of affinity purified SMN macromolecular complexes. Immunofluorescence microscopy of HeLa cells showed GEMIN6 staining in both the cytoplasm and the nucleus with a high concentration in gems.
