Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate CHRM2 in samples. An antibody specific for CHRM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCHRM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHRM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHRM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

In the heart, this contributes to a decreased heart rate.The M2 muscarinic receptors are located in the heart, where they act to slow the heart rate down to normal sinus rhythm after stimulatory actions of the sympathetic nervous system, by slowing the speed of depolarization. They also reduce contractile forces of the atrial cardiac muscle, and reduce conduction velocity of the atrioventricular node (AV node). However, they have no effect on the contractile forces of the ventricular muscle.
