Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate MPZ-Ab-IgG in samples. An antibody specific for MPZ-Ab-IgG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMPZ-Ab-IgG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPZ-Ab-IgG is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPZ-Ab-IgG bound in the initial step. The color development is stopped and the intensity of the color is measured.

(1991) found that the Mpz gene in the mouse, like that in the rat, contains 6 exons that span about 7 kb of genomic DNA.Myelin protein-zero is the major structural protein of peripheral myelin, accounting for more than 50% of the protein present in the sheath of peripheral nerves. Expression of the MPZ gene is restricted to Schwann cells; MPZ is not found in the CNS. An integral membrane glycoprotein of 28 kD, MPZ is thought to link adjacent lamellae and thereby stabilize the myelin assembly. The other 3 major components of myelin are myelin basic protein (MBP), myelin proteolipid protein (PLP1), and myelin-associated glycoprotein (MAG).
