Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate GHRP in samples. An antibody specific for GHRP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGHRP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GHRP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GHRP bound in the initial step. The color development is stopped and the intensity of the color is measured.

The pathway activated by binding of Ghrelin to the Growth hormone secretagogue receptor, GHSR1a, regulates the activation of the downstream mitogen-activated protein kinase, Akt, nitric oxide synthase, and AMPK cascades in different cellular systems. One of the important features of GHSR1a displays constitutive activity possessing basal activity in the absence of an agonist, resulting in a high degree of receptor internalization as well as of signaling activity.
