Biotinylated proteins including primary and secondary antibodies are widely used in protein purification and immunoassays. Due to its high binding affinity to streptavidin, biotinylated proteins are often used with streptavidin conjugates such as streptavidin-fluorescent dyes and streptavidin-HRP to amply the signals and increase detection sensitivity. Streptavidin-polymer-HRP combine the specificity of streptavidin-biotin interaction and signal amplification through multiple conjugations of HRP to a hydrophilic polymer backbone. The applications of this highly sensitive detection of biotinylated proteins include Western blotting, ELISA, immunoprecipitation, immunocytochemistry and immunohistochemistry.
